Psoriatic Arthritis model / Pathology Overview

Psoriatic Arthritis model is important as research model as Psoriatic Arthritis is a type of arthritis that affects some people who have psoriasis, a disease that causes red patches of skin topped with silvery scales lesions. Most people develop psoriasis years before being diagnosed with Psoriatic Arthritis. However, for some, joint problems appear before or at the same time as skin lesions.

Psoriatic Arthritis is characterized by joint pain, stiffness, and swelling. They can affect any part of the body, including fingertips and spine, and their severity can range from mild to severe. Disease flares and remissions can occur in both psoriasis and Psoriatic arthritis.

Psoriatic Arthritis has no known cure. The goal of treatment is to alleviate symptoms while preventing joint damage. Psoriatic Arthritis can be disabling if not treated.

Mannan-induced Psoriatic Arthritis mouse model

Model Induction:

Single IP of Mannan in DBA mice strain with 8-days in vivo follow-up

Macroscopical Scoring / Clinical / Daily observations:

  • Dermatogical endpoints:
    • Erythema (red skin appearance)
      • Scoring (by operator)
      • Colorimetric evaluation (operator independent measurement)
    • Scaling (dry skin appearance) / Scoring
    • Ear thickness / edema (calipering)

    • Arthritis scoring hindlimb/toes
    • Hindlimb thickness / edema (calipering)

In vivo imaging / Intermediate times:

  • Optical Coherence Tomography / OCT on ears/back skin
  • Thermography / FLIR on ears/hindlimbs

At termination:

  • Hindlimb and ears pictures
  • Collection of hindlimb and ears for histology
    • HES staining
    • Other stainings on request

  • Betamethasone / PO daily / Therapeutic
  • Ciclosporine / PO daily / Prophylactic
  • Methotrexate / IP / Prophylactic
